Biological Background: NTAN1 Function and Localization
- NTAN1 (also known as PNAA or PNAD) is a protein N-terminal asparagine amidohydrolase that catalyzes the deamidation of N-terminal asparagine residues to aspartate.
- This enzyme is essential for the N-end rule pathway, enabling the ubiquitin-dependent degradation of proteins that start with Met-Asn by facilitating subsequent arginylation and polyubiquitination.
- NTAN1 specifically acts on proteins with an exposed N-terminal asparagine after removal of the initiator methionine; it does not modify internal or C-terminal asparagine residues or any glutamine residues.
- The protein localizes to the cytoplasm, consistent with its role in cytosolic protein quality control.
- NTAN1 is a member of the hydrolase family and has a reference proteome status; its three-dimensional structure has been experimentally determined.
- It contributes to proteostasis by regulating the turnover of proteins with destabilizing N-terminal residues.
Experimental Guidance and Technical Tips
- The immunogen spans almost the full-length NTAN1 protein (residues 1–310), which may result in recognition of multiple epitopes and could be suitable for detecting endogenous NTAN1 in various sample types.
- When performing Western blotting, a band around 35 kDa is predicted; consider using appropriate positive controls (e.g., human, mouse, or rat cell lysates) and validate detection in your specific experimental system.
- The reported cross-reactivity with human, mouse, and rat suggests conserved epitopes, making this antibody a potential tool for cross-species studies; however, verify performance in your target species.
- For ELISA, the antibody may be used as a detection reagent; optimization of coating antigen concentration and antibody dilution is recommended to achieve optimal signal-to-noise ratios.
