  • NPHP3 (Nephrocystin-3) is a protein that contains TPR repeats and coiled coil domains, functioning as a key regulator in ciliary biology and Wnt signaling pathways.
  • It localizes specifically to the cilium, a microtubule-based organelle, where it is essential for normal ciliary development and function.
  • NPHP3 inhibits the canonical Wnt signaling cascade by suppressing Disheveled-1 activity, and may also modulate non-canonical Wnt/planar cell polarity pathways, acting as a molecular switch between these pathways.
  • The protein is required for convergent extension cell movements during development, a process mediated by non-canonical Wnt signaling.
  • NPHP3 is widely expressed at low levels, with relatively higher expression in heart, placenta, liver, skeletal muscle, kidney, and pancreas, and very low in brain and lung.
