Biological Background: Nephrocystin-1 Function and Localization
- Nephrocystin-1 (NPHP1) is encoded by the NPHP1 gene and is also known as juvenile nephronophthisis 1 protein.
- It plays a role in epithelial cell polarity and the organization of apical junctions in kidney cells, interacting with proteins such as BCAR1, NPHP4, and RPGRIP1L.
- NPHP1 is involved in the regulation of intraflagellar transport (IFT) during cilia assembly, influencing the movement of IFT proteins like IFT88 and WDR19 in photoreceptor cilia.
- It localizes to cell junctions, including tight junctions and adherens junctions, and is found within the cilium axoneme and cytoplasm.
- NPHP1 is required for normal retina development and spermatogenesis.
- Tissue expression is widespread, with highest levels in pituitary gland, spinal cord, thyroid, testis, skeletal muscle, lymph node, and trachea; lower in heart, kidney, and pancreas. Related references: PMID:16308564 PMID:18477472
