Biological Background: NOX5 Function and Localization
- NOX5 is a calcium-dependent NADPH oxidase that generates superoxide (O2•−) from molecular oxygen using NADPH as an electron donor, contributing to reactive oxygen species (ROS) signaling. Related references: PMID:11483596, PMID:14982937, PMID:17275676, PMID:12686516
- An isoform lacking calcium-binding EF-hand domains exhibits calcium-independent oxidase activity, though its activity is debated; it may undergo rapid degradation. Related references: PMID:17275676, PMID:36653838, PMID:22427510
- In endothelial cells, NOX5 mediates thrombin-induced ROS production, proliferation, and angiogenesis, highlighting its role in vascular biology. Related references: PMID:17275676
- NOX5 also synthesizes the calcium-mobilizing second messenger NAADP from NAADPH, impacting T cell activation. Related references: PMID:34784249
- Subcellularly, NOX5 localizes to the endoplasmic reticulum and cell membrane.
- It is primarily expressed in testis (pachytene spermatocytes), spleen, lymph nodes, and endothelial cells, with additional expression in ovary, placenta, pancreas, and cardiac fibroblasts.
- The protein contains FAD- and calcium-binding domains, characteristic of the NADPH oxidase family, and features transmembrane helices.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 661–765 (C-terminal region) of human NOX5; the antibody is expected to recognize this region. Consider validating specificity using overexpression or knockdown systems.
- For Western blot, anticipate a band near 86 kDa; additional bands may appear due to isoforms or post-translational modifications; validate in the relevant sample system.
- In immunofluorescence/immunocytochemistry, optimize fixation and permeabilization methods as NOX5 localizes to both endoplasmic reticulum and plasma membrane.
- For ELISA, the antibody may be used for detection; appropriate positive and negative controls are recommended.
- Cross-reactivity has been reported for human only; verify reactivity in other species if needed.
