Biological Background: NLRX1 Function and Localization

  • NLRX1 (NOD5, NOD9, NOD26, DLNB26, CLR11.3) is a member of the NLR (nucleotide-binding domain and leucine-rich repeat containing) family, characterized by an N-terminal mitochondrial targeting sequence (transit peptide), a central NACHT domain with ATP-binding activity, and C-terminal leucine-rich repeats (LRRs).
  • Negatively regulates the virus-induced RLH-MAVS interaction, thereby inhibiting MAVS-mediated antiviral signaling (PubMed:18200010). Related references: PMID:18200010
  • Promotes autophagy by interacting with TUFM and subsequently recruiting ATG5 and ATG12 (PubMed:22749352). Related references: PMID:22749352
  • Regulates MAVS-dependent NLRP3 inflammasome activation to attenuate apoptosis (PubMed:27393910). Related references: PMID:27393910
  • Enhances NF-kappa-B and JNK signaling through the production of reactive oxygen species (PubMed:18219313). Related references: PMID:18219313
  • Exhibits sex-dependent regulation of viral-mediated inflammation and energy metabolism, potentially contributing to sex differences observed in infectious diseases (By similarity).
  • Localizes to the mitochondrial outer membrane.
  • Ubiquitously expressed, with strongest levels in mammary gland, heart, and muscle; detected in HeLa, HEK293T, THP-1, HL-60, Raji, and Jurkat cell lines (at protein level).

Experimental Guidance and Technical Tips

  • For Western blot, target protein runs at approximately 108kDa; consider verifying band specificity with knockdown controls and using appropriate protein ladders.
  • In immunoprecipitation, test antibody suitability with endogenous NLRX1 from relevant cell lines (e.g., HeLa, HEK293T) known to express the protein.
  • The immunogen corresponds to a recombinant fragment (aa 87-360); multiple isoforms or splice variants may be detected; validate in your experimental system.
  • Cross-reactivity with mouse and rat is reported; for non-human samples, verify recognition using appropriate tissue or cell lysates.
