Biological Background: NLRP1 Function and Localization

  • NLRP1 (NACHT, LRR and PYD domains-containing protein 1) is a multidomain protein also known as CARD7, DEFCAP, NAC, and NALP1. It serves as the sensor component of the NLRP1 inflammasome, a supramolecular complex that assembles in the cytosol in response to pathogen-associated signals and mediates pyroptosis and inflammatory cytokine release.   • Upon activation, NLRP1 recruits pro-caspase-1 (proCASP1) via the adaptor PYCARD/ASC, triggering CASP1 autoproteolysis. Active CASP1 cleaves pro-IL1B, pro-IL18, and gasdermin-D (GSDMD), leading to cytokine secretion and pyroptotic cell death. Experimental Guidance and Technical Tips

  • For Western blotting, NLRP1 has a predicted molecular weight of ~166 kDa. Use a low-percentage polyacrylamide gel (e.g., 6–8%) and extended wet transfer times (overnight at low voltage) to ensure efficient transfer of this large protein. Include a high-molecular-weight prestained marker as a reference.
  • For IHC-P, antigen retrieval (citrate or EDTA buffer, heat-induced) is recommended. Validate the antibody in your tissue of interest (human, mouse, or rat) with appropriate positive and negative controls, and titrate to minimize background.
