Biological Background: NF-κB2 Function and Localization
- NF-κB2 encodes the p100 precursor and its processed p52 subunit, a component of the NF-κB transcription factor family.
- It forms homo- or heterodimers with Rel proteins (RELA, RELB, REL) to regulate genes involved in immune response, inflammation, cell growth, and apoptosis.
- In the non-canonical NF-κB pathway, p100 undergoes proteasomal processing to p52, leading to formation of the RelB-p52 activator complex.
- p52-p52 homodimers act as transcriptional repressors, while p52-RELA and p52-RELB complexes can activate target genes.
- The protein localizes to both the cytoplasm and nucleus; cytoplasmic p100 retains associated NF-κB proteins, while p52 translocates to the nucleus upon activation.
- Post-translational modifications include phosphorylation, ubiquitination, and processing events that regulate its activity (UniProt keywords: Phosphoprotein, Isopeptide bond, Ubl conjugation).
- NF-κB2 has been implicated in circadian clock regulation by repressing CLOCK-BMAL1 activity together with RELB.
- Genetic rearrangements and mutations in NFKB2 are associated with immunodeficiencies such as CVID10.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 690-899 of human NF-κB2 (p100 C-terminal region), which may recognize both the p100 precursor and the p52 processed form.
- For Western blot (WB), a band around 97 kDa (p100) and a smaller band around 52 kDa (p52) may be observed, depending on cellular processing.
- Consider using appropriate cell lysates or stimuli that activate the non-canonical NF-κB pathway to assess p100 processing to p52.
- For immunofluorescence/immunocytochemistry (IF/ICC), fixation and permeabilization should be optimized to detect both cytoplasmic and nuclear pools.
- Cross-reactivity with mouse samples has been reported; validate in the relevant sample system.
- ELISA may be suitable for detecting recombinant or endogenous NF-κB2 protein; titration of antibody concentration is recommended.
