Biological Background: NF-kB p65/RelA Function and Localization

  • NF-kB p65 (RELA) is the p65 subunit of the pleiotropic NF-kappa-B transcription factor complex. It belongs to the NF-kappa-B/Rel protein family, which also includes RELB, NFKB1/p105/p50, REL, and NFKB2/p52.
  • RELA forms homo- or heterodimers with other family members; the RELA-NFKB1 heterodimer is the most abundant. These dimers bind specific DNA kappa-B sites to activate transcription of target genes involved in inflammation, immunity, cell growth, and apoptosis.
  • The protein shuttles between the cytoplasm and nucleus. In unstimulated cells, it is retained in the cytoplasm via interaction with I-kappa-B inhibitor proteins. Upon stimulation, I-kappa-B is phosphorylated and degraded, freeing RELA to translocate to the nucleus and regulate gene expression.
