Biological Background: Neurofilament H Function and Localization
- Neurofilament H (NFH), also known as 200 kDa neurofilament protein or neurofilament triplet H protein, is encoded by the NEFH gene and belongs to the intermediate filament family.
- NEFH is a key component of the neuronal cytoskeleton and, together with NEFL and NEFM, maintains neuronal caliber; it has a unique and essential role in mature axons that cannot be fully compensated by the two smaller neurofilament proteins.
- It may also cooperate with the neuronal intermediate filament proteins peripherin (PRPH) and internexin (INA) to form robust filamentous networks in neurons.
- Subcellular localization is predominantly cytoplasmic, with enrichment in the cytoskeleton, cell projections, and particularly axons.
- The protein is subject to extensive post-translational modifications, including phosphorylation, and is implicated in several neurodegenerative diseases such as Charcot-Marie-Tooth disease type 2CC and amyotrophic lateral sclerosis.
- UniProt lists the molecular weight as approximately 112 kDa (111838 Da), which is consistent with the commonly reported size of the full-length protein.
Experimental Guidance and Technical Tips
- Given the immunogen spans the C-terminal tail region (aa 873–1020), this antibody is expected to detect full-length NEFH and may not cross-react with truncated isoforms lacking this region. Consider validating in your sample system.
- For Western blotting, a band around 112 kDa is anticipated. Use appropriate protein loading controls and sample preparation to prevent degradation of this large protein.
- In immunofluorescence (IF-P) and immunohistochemistry (IHC-P), optimize fixation and permeabilization protocols to preserve the filamentous structure; strong axonal staining is expected in neuronal tissues.
