Biological Background: NEURL1B Function and Localization
- NEURL1B encodes an E3 ubiquitin-protein ligase, a member of the neuralized family of RING-type E3 ligases, and is also known as Neuralized-2 (NEUR2), NEURL3, or RNF67B.
- The protein functions in the Notch signaling pathway by modulating the stability and activity of several Notch ligands, thereby influencing downstream signaling events.
- NEURL1B is localized to the cytoplasm, consistent with its role in substrate ubiquitination and protein turnover.
- Tissue expression is highest in the brain, prostate, and small intestine, with higher levels in fetal brain compared to adult; in adult brain, strong expression is observed in the olfactory system, cerebellar cortex, optic nerve, and frontal lobe.
- The protein contains a RING-type zinc finger and NEUZ domains, indicative of its E3 ubiquitin ligase activity and metal-binding properties.
- Multiple isoforms exist due to alternative splicing, and the protein is subject to phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 270–440 of human NEURL1B (NP_001136123.1). This region may recognize multiple isoforms if the epitope is conserved; validate isoform detection in your system.
- Recommended applications include Western blot (WB), immunohistochemistry-paraffin (IHC-P), and ELISA. Optimize antibody dilutions for each assay using appropriate positive and negative controls.
- For WB, the predicted molecular weight of NEURL1B is approximately 59 kDa; confirm band specificity against lysates from tissues with high expression (e.g., brain).
- As a polyclonal antibody, it may exhibit batch-to-batch variation; consider lot-specific validation for critical experiments.
