Biological Background: NDUFA4 Function and Localization
- NDUFA4, also known as COXFA4, CI-MLRQ, or MLRQ, is a nuclear-encoded subunit of cytochrome c oxidase (complex IV) and was historically annotated as a subunit of NADH dehydrogenase (complex I). It is encoded by the COXFA4 gene in humans.
- NDUFA4 is an integral component of cytochrome c oxidase, the terminal enzyme of the mitochondrial electron transport chain, where it is required for complex IV maintenance and the efficient reduction of oxygen to water. Related references: PMID:22902835
- The protein localizes to the mitochondrial inner membrane and contains a transmembrane helix, positioning it within the lipid bilayer to participate in electron transfer.
- Post-translational modifications include acetylation and phosphorylation, which may regulate its activity or assembly into the respiratory supercomplex.
- Mutations in NDUFA4 have been associated with Leigh syndrome, a severe primary mitochondrial disease characterized by progressive neurodegeneration.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–81), which is predicted to produce antibodies recognizing full-length endogenous NDUFA4 across human, mouse, and rat.
- For Western blot, a specific band near 9 kDa is expected in mitochondrial-enriched lysates; validate in the relevant sample system using appropriate loading controls.
- In IHC-P and IF/ICC, consider using antigen retrieval and permeabilization protocols optimized for mitochondrial membrane proteins to enhance detection.
- ELISA performance may rely on the recombinant immunogen or native protein; titer and specificity should be confirmed with the appropriate antigen format.
