Biological Background: N-acetyl-D-glucosamine kinase Function and Localization
- N-acetyl-D-glucosamine kinase (NAGK, also known as GlcNAc kinase, muramyl dipeptide kinase, or N-acetyl-D-mannosamine kinase) is a ubiquitously expressed enzyme encoded by the NAGK gene.
- Catalyzes the phosphorylation of N-acetylglucosamine (GlcNAc) to GlcNAc 6-phosphate, playing a central role in recycling GlcNAc from lysosomal degradation and dietary sources. Related references: PMID:22692205
- Involved in the N-glycolylneuraminic acid (Neu5Gc) degradation pathway, processing Neu5Gc from food, despite humans lacking the ability to synthesize Neu5Gc. Related references: PMID:22692205
- Exhibits N-acetylmannosamine (ManNAc) kinase activity, contributing to sialic acid metabolism (by similarity).
- Functions in innate immunity by phosphorylating muramyl dipeptide (MDP) to 6-O-phospho-MDP, a potent NOD2 ligand that triggers antibacterial responses. Related references: PMID:36002575
- Post-translationally regulated by acetylation and phosphorylation, with at least one alternatively spliced isoform reported.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 50–210 of human NAGK, a region that may be conserved across species; consider verifying sequence homology for your target species to ensure reactivity.
- For Western blotting (WB), a band around 37 kDa is expected; validate in your sample system and consider using positive controls from human, mouse, or rat lysates.
- ELISA performance may depend on coating conditions and detection format; pilot testing with recombinant protein or peptide controls is recommended.
- Due to NAGK’s ubiquitous expression, inclusion of appropriate negative controls (e.g., knockout lysates or siRNA-treated samples) can help confirm specificity.
