Biological Background: MYO10 Function and Localization

  • MYO10 encodes unconventional myosin-X, also known as unconventional myosin-10, a member of the myosin superfamily of actin-based motor proteins with ATPase activity.
  • It functions as a plus end-directed motor that binds to actin filaments and bundles, moving with higher velocity and taking larger steps on actin bundles than on single actin filaments. Related references: PMID:27580874
  • MYO10 plays a role in cell shape, spreading, and adhesion, and stimulates the formation and elongation of filopodia. In hippocampal neurons, it induces dendritic filopodia by trafficking VASP to filopodial tips to promote actin elongation.
  • The protein localizes to the cytoplasm, cytosol, cell projections such as lamellipodia, ruffles, and filopodia tips, as well as the cell cortex and filopodium membrane.
  • It is ubiquitously expressed and regulated by alternative promoter usage and splicing, giving rise to isoforms; notably, an isoform acts as a dominant-negative regulator of isoform 1, suppressing filopodia induction and axon outgrowth.
  • Keywords include actin-binding, calmodulin-binding, phosphoprotein, and acetylation, indicating post-translational modifications.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 845–944 of human MYO10, a region that may be conserved across isoforms; consider verifying isoform specificity in your experimental system.
  • For Western blot, target protein weight is approximately 237 kDa; use appropriate gel percentage (e.g., 6–8% SDS-PAGE) and ensure efficient transfer of high-molecular-weight proteins (e.g., extended transfer time or wet transfer).
  • The antibody is validated for WB and ELISA; it may also be suitable for other applications such as immunoprecipitation or immunofluorescence, but these require independent optimization.
  • Cross-reactivity with human and mouse is reported; for other species, check sequence homology of the immunogen region.
