Biological Background: mTOR Function and Localization
- mTOR (Mechanistic Target of Rapamycin) is a serine/threonine protein kinase that serves as a central regulator of cellular metabolism, growth, and survival, integrating signals from hormones, growth factors, nutrients, energy, and stress. Related references: PMID:12087098 PMID:12150925 PMID:12150926
- It functions within two distinct complexes: mTORC1 and mTORC2. mTORC1 is nutrient-sensitive and promotes anabolic processes like protein, lipid, and nucleotide synthesis, while mTORC2 is nutrient-insensitive and regulates proliferation, survival, and cytoskeletal organization by activating AGC kinases such as AKT, PKC, and SGK1. Related references: PMID:15268862 PMID:15467718 PMID:18497260 PMID:21376236 PMID:24670654
- mTORC1 directly phosphorylates key translational regulators including EIF4EBP1 (4E-BP1) and RPS6KB1 (S6K), thereby enhancing mRNA translation and ribosome biogenesis. Related references: PMID:12087098 PMID:12150925 PMID:12150926 PMID:24403073 PMID:29236692
- mTORC1 also regulates lipid synthesis via SREBF1 and suppresses autophagy by phosphorylating ULK1 and other autophagy-related proteins. Related references: PMID:23426360 PMID:32561715 PMID:20537536
- mTORC2-mediated phosphorylation of AKT at Ser473 is critical for full AKT activation, influencing cell survival, metabolism, and migration. Related references: PMID:15268862 PMID:15467718 PMID:21376236 PMID:24670654
- Subcellularly, mTOR dynamically localizes to lysosomes, endoplasmic reticulum, Golgi apparatus, mitochondria, nucleus, and cytoplasmic vesicles in response to nutrient and growth factor cues.
- mTOR is widely expressed across tissues, with the highest levels observed in the testis.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to the N-terminal region (amino acids 1-300) of human mTOR. This sequence is conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blot (WB), expect a band at approximately 289 kDa. Use appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
- In immunoprecipitation (IP), the antibody may pull down mTOR and its associated complex components; validate under native conditions.
- For immunofluorescence (IF/ICC), staining may appear in cytoplasmic, lysosomal, and membrane compartments depending on cell type and culture conditions. Include appropriate controls.
- Optimal dilutions and conditions should be determined empirically for each application and sample type.
