Biological Background: MTF2 Function and Localization
- MTF2 (Metal-response element-binding transcription factor 2), also known as Polycomb-like protein 2 (PCL2) or M96, is a Polycomb group (PcG) protein that functions as a chromatin regulator with zinc-finger and DNA-binding domains.
- As a reader of histone modifications, MTF2 specifically binds trimethylated histone H3 at lysine 36 (H3K36me3) and recruits the PRC2 complex, thereby enhancing its H3K27me3 methyltransferase activity. Related references: PMID:23142980, PMID:23228662, PMID:31959557
- MTF2 is crucial for regulating transcriptional networks during embryonic stem cell self-renewal and differentiation.
- It promotes recruitment of the PRC2 complex to the inactive X chromosome in differentiating XX ES cells and to target genes in undifferentiated ES cells.
- MTF2 is required to repress Hox genes by enhancing H3K27me3 methylation of the PRC2 complex at these developmental loci.
- In certain contexts, MTF2 may act as an inhibitor of PRC2 activity, activating the CDKN2A gene and promoting cellular senescence.
- The protein localizes to the nucleus and is also capable of binding to the metal-regulating element (MRE) of the MT1A gene promoter, suggesting a role in metal response.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–190) of human MTF2, which may contain conserved domains; cross-reactivity with mouse and rat is confirmed, but validation in these species is recommended.
- For Western blotting, nuclear protein extracts are recommended to enrich the target protein, and a band around 67 kDa should be observed.
- For ELISA applications, the antibody may be used to detect recombinant MTF2 or endogenous protein in cell lysates, but optimization of coating and detection conditions is advised.
