Biological Background: CD3 Function and Localization
- CD3E is a subunit of the TCR-CD3 complex essential for adaptive immunity and transmits activation signals across the T-cell membrane upon antigen recognition. Related references: PMID:15294938 PMID:15546002 PMID:2470098 PMID:40592325
- The cytoplasmic domain of CD3E contains immunoreceptor tyrosine-based activation motifs (ITAMs) that are phosphorylated by LCK and FYN Src kinases, initiating downstream signaling pathways. Related references: PMID:2470098 PMID:40592325
- CD3E also regulates TCR internalization and cell surface down-regulation via endocytosis sequences in its cytosolic tail. Related references: PMID:10384095 PMID:26507128

- CD3E is required for correct T-cell development (by similarity) and localizes to the cell membrane as a single-pass type I membrane protein.
