Biological Background: Alpha-(1,3)-fucosyltransferase 4 (FUT4) Function and Localization
- Alpha-(1,3)-fucosyltransferase 4 (FUT4) is a Golgi-resident glycosyltransferase that synthesizes the Lewis X (LeX) glycan determinant (CD15) by transferring fucose in alpha(1,3)-linkage to N-acetylglucosamine (GlcNAc) on type 2 lactosamine chains. - CD15/LeX is a key cell-surface glycoepitope involved in cell adhesion, migration, and immune cell recognition, particularly through interactions with selectins (SELE, SELL, SELP) during leukocyte trafficking and homing.
- FUT4 works together with FUT7 to decorate selectin ligands with fucosylated glycans, a critical step in lymphocyte recirculation and inflammatory responses.
