Biological Background: Integrin alpha-M Function and Localization
- Integrin alpha-M (ITGAM, CD11b) pairs with beta-2 integrin (ITGB2/CD18) to form macrophage-1 antigen (Mac-1) or complement receptor 3 (CR3), which mediates adhesive interactions of monocytes, macrophages, and granulocytes and facilitates uptake of complement-coated particles and pathogens. Related references: PMID:20008295 PMID:9558116
- It serves as a receptor for the iC3b fragment of complement component 3, likely recognizing the R-G-D peptide, and also binds fibrinogen and factor X via specific gamma chain peptides.
- Regulates neutrophil migration, contributing to innate immune responses. Related references: PMID:28807980
- In association with ITGB2, it is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils. Related references: PMID:21193407
- Recognizes intercellular adhesion molecule 1 (ICAM1), enabling adhesion between stimulated neutrophils and endothelial cells. Related references: PMID:1980124
- May also regulate phagocytosis-induced apoptosis in extravasated neutrophils, contribute to mast cell development, and control microglial superoxide production during brain development (by similarity).
- Subcellular localization is primarily the cell membrane and membrane rafts.
- Predominantly expressed in monocytes and granulocytes, with confirmed protein expression in neutrophils.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide derived from human CD11b; consider verifying that the antibody recognizes the native conformation of integrin alpha-M on live cells by flow cytometry.
- For flow cytometry applications, optimize cell preparation, antibody titration, and appropriate isotype controls to ensure specific staining.
- Use human peripheral blood monocytes or granulocytes as positive controls, and lymphocytes (which lack CD11b) as negative controls.
- Fixation and permeabilization protocols may affect epitope accessibility; validate in the relevant sample system.
