Biological Background: Integrin alpha-M Function and Localization
- Protein identity: Integrin alpha-M (CD11b) is a transmembrane glycoprotein and a member of the integrin alpha chain family, also known as CR-3 alpha chain or MAC-1 subunit alpha.
- Adhesive and receptor functions: ITGAM/ITGB2 integrin mediates adhesive interactions of myeloid cells and serves as a receptor for iC3b, fibrinogen, factor X, and ICAM1, facilitating phagocytosis of complement-coated particles and neutrophil adhesion. Related references: PMID:20008295, PMID:9558116
- Role in neutrophil activation: In association with ITGB2/CD18, required for CD177-PRTN3-mediated activation of TNF-primed neutrophils. Related references: PMID:21193407
- Regulation of neutrophil migration: Contributes to the control of neutrophil trafficking during immune responses. Related references: PMID:28807980
- Subcellular localization: Localizes to the cell membrane and is enriched in membrane rafts, consistent with its role in adhesion and signaling.
- Tissue specificity: Predominantly expressed on monocytes and granulocytes, including neutrophils. Related references: PMID:1346576, PMID:21193407
- Structural features: Contains disulfide bonds and multiple glycosylation sites; activity depends on divalent cations such as calcium and magnesium.
Experimental Guidance and Technical Tips
- For flow cytometry, consider titrating the antibody in the relevant sample system to determine the optimal staining concentration.
- Use human monocytes or granulocyte populations as positive controls to verify specificity and performance.
- Ensure appropriate isotype controls and viability dyes are included to discriminate live, CD11b-positive cells.
- This antibody is validated for flow cytometry; if adapting to other applications, preliminary testing is recommended.
