Biological Background: NONO/p54nrb Function and Localization

NONO (Non-POU domain-containing octamer-binding protein), also known as p54nrb, is a multifunctional DNA- and RNA-binding protein involved in diverse nuclear processes. It binds both double- and single-stranded DNA and RNA, participates in pre-mRNA splicing likely as a heterodimer with SFPQ, and interacts with U5 snRNA. NONO is essential for paraspeckle formation together with PSPC1, and it contributes to DNA repair via non-homologous end joining (NHEJ) by stabilizing DNA ends with the Ku70/Ku80 dimer.

In transcriptional regulation, NONO forms a complex with SFPQ and NR5A1 to modulate CYP17 promoter activity. It also regulates the circadian clock by repressing CLOCK-BMAL1 transactivation, influences GABAergic synapse organization, and promotes neuronal differentiation by recruiting TET1 to genomic loci.

NONO plays a critical role in innate immunity by assembling into the HDP-RNP complex, which serves as a platform for IRF3 phosphorylation, thereby activating the cGAS-STING pathway in response to DNA virus infection. During HIV-2 infection, NONO interacts with the viral capsid protein p24 to facilitate viral DNA sensing by cGAS.

Subcellularly, NONO is predominantly nuclear, concentrating in nucleoli, nuclear speckles, and punctate paraspeckles adjacent to splicing speckles.

NONO is widely expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney, pancreas, and various breast tumor cell lines. The protein undergoes extensive post-translational modifications, including acetylation, phosphorylation, and methylation (e.g., monomethylation at R456). This antibody specifically recognizes the mono-methylated form at arginines 357 and 365.

  • This rabbit polyclonal antibody was raised against a synthetic peptide carrying monomethylation at R357 and R365 of human NONO/p54nrb. For Western blot, a band around 54 kDa is expected.
