Biological Background: Histone H4 Function and Localization
Histone H4 is one of the core histone proteins that, together with H2A, H2B, and H3, forms the nucleosome—the fundamental repeating unit of chromatin. Each nucleosome consists of 147 base pairs of DNA wrapped around an octamer of two copies each of the four core histones. This structure compacts the genomic DNA, but also serves as a dynamic platform for regulating DNA accessibility. Histone H4 plays a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. These processes are tightly controlled by an extensive array of post-translational modifications on histone tails, collectively referred to as the "histone code," which orchestrate nucleosome remodeling and the recruitment of effector proteins.
Histone H4 is predominantly localized to the nucleus, where it associates with chromosomes throughout the cell cycle. During spermatogenesis, acetylated H4 shows specific nuclear localization patterns in elongating spermatids.
The N-terminal tail of Histone H4 is subject to a wide variety of post-translational modifications, including acetylation, methylation, phosphorylation, citrullination, and many others. Methylation of arginine residues is a key regulatory mark. MonoMethylation at arginine 3 (H4R3me1) is catalyzed by protein arginine methyltransferases and has been implicated in transcriptional regulation and the establishment of chromatin states.

Given its high conservation across species, Histone H4 sequence and modification patterns are broadly preserved from yeast to human, supporting the wide cross-reactivity of this antibody.
Experimental Guidance and Technical Tips
- Since this antibody was raised against a synthetic monomethylated peptide corresponding to residues around R3 of human Histone H4, it is expected to specifically recognize the monomethylated form of H4R3. Researchers should consider validating specificity using peptide competition assays or with cell lines treated with methyltransferase inhibitors.
- For immunofluorescence (IF/ICC), standard fixation with 4% paraformaldehyde followed by permeabilization with 0.1–0.5% Triton X-100 is typically compatible with histone detection. Nuclear counterstaining (e.g., DAPI) is recommended to confirm nuclear localization of the signal.
- In ELISA applications, the antibody can be used to detect synthetic monomethylated H4R3 peptides or histone extracts from cells. Consider optimizing coating buffer and antibody dilution for your specific assay system.
- The antibody is predicted to cross-react with Histone H4 from human, mouse, and rat, as well as a wide range of other species due to high sequence conservation. However, cross-reactivity should be experimentally validated in the relevant sample system.
