Biological Background: Histone H4 Function and Localization
Histone H4 is a core component of the nucleosome, wrapping and compacting DNA into chromatin. This limits DNA accessibility and plays a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is regulated through a complex set of post-translational modifications, collectively termed the histone code, and nucleosome remodeling. Related references: UniProt P62805
Histone H4 localizes to the nucleus and is a structural component of chromosomes. Acetylated H4 is detected in the nucleus of step 11 spermatids. Related references: UniProt P62805
Lysine 20 of histone H4 (corresponding to position 21 in the full-length protein) is subject to mono-, di-, and tri-methylation. Monomethylation at H4K20 has been implicated in chromatin compaction, heterochromatin formation, and DNA damage response. These modifications add functional diversity to the histone code. Histone H4 is highly conserved across species, with a wide range of other post-translational modifications including acetylation, citrullination, phosphorylation, and ubiquitination at multiple residues. This conservation makes H4 a valuable target for studying epigenetic regulation in diverse model organisms. Related references: UniProt P62805
Experimental Guidance and Technical Tips
- The immunogen is a synthetic monomethylated peptide around K20; consider verifying antibody specificity by peptide competition ELISA using unmodified, mono-, di-, and tri-methylated K20 peptides.
- For Western blot, acid extraction of histones or nuclear fractionation is recommended to enrich for histone H4. The predicted molecular weight is approximately 11 kDa; use a 15% SDS-PAGE gel or gradient gel for optimal separation of low-molecular-weight histones.
- Given the broad predicted cross-reactivity (human, mouse, rat, and others), validate the antibody in the relevant sample system, as signal intensity may vary depending on endogenous methylation levels.
- An ELISA assay can be used to quantify relative monomethyl-H4K20 levels using standardized histone preparations.
