Biological Background: Histone H3 (H3.1/H3.1t) Function and Localization

  • Histone H3 is a core component of the nucleosome, wrapping and compacting DNA into chromatin. This limits DNA accessibility and plays a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is controlled through the "histone code" — a complex set of post-translational modifications (PTMs) and nucleosome remodeling.
  • The protein localizes to the nucleus and chromosomes.
  • Two major isoforms are relevant: H3.1 (encoded by multiple genes, e.g., H3C1, H3C2, etc., UniProt P68431) is expressed in most somatic cells, whereas H3.1t (H3-4, HIST3H3, UniProt Q16695) is testis-specific.
  • The N-terminal tail of histone H3 is subject to numerous PTMs, including acetylation, methylation, phosphorylation, citrullination, and others. Specifically, arginine 2 (R2) can be methylated. Asymmetric dimethylation at this site is catalyzed by PRMT6 and is associated with transcriptional regulation. Monomethylation at R2 is an intermediate step and may have distinct functional roles. Experimental Guidance and Technical Tips

  • The immunogen is a synthetic monomethylated peptide around R2 of human histone H3. When performing western blot (WB), consider using nuclear extracts or histone-enriched preparations to increase signal. The predicted molecular weight is ~15 kDa, but bands may appear slightly higher due to PTMs or protein interactions.
  • For immunofluorescence/immunocytochemistry (IF/ICC), fixation and permeabilization conditions may need optimization. Since histone H3 is nuclear, permeabilization with Triton X-100 or methanol is recommended. Validate the staining pattern against known nuclear markers.
  • In ELISA, consider using a methyl-peptide as the coating antigen to confirm specificity for monomethyl R2 versus unmodified or dimethylated forms.
  • Species cross-reactivity is predicted to be wide (human, mouse, rat, and others), but it is advisable to verify reactivity in the specific sample system of interest, especially for non-mammalian species.
