Both share the same fundamental role in chromatin structure.

Lysine 4 of histone H3 (H3K4) is a major site for diverse post-translational modifications, including mono-, di-, and trimethylation, acetylation, and others. Monomethylation of H3K4 (H3K4me1) is typically associated with enhancer regions and active gene transcription. Experimental Guidance and Technical Tips

  • This antibody is specific for monomethylated H3K4. For optimal results, include positive and negative control samples (e.g., peptides with unmodified, mono-, di-, or tri-methylated K4) to confirm specificity.
  • Western Blot (WB): Use nuclear extracts or whole cell lysates. A band at approximately 16 kDa is expected. Determine the optimal primary antibody dilution empirically (e.g., starting range 1:500–1:2000) and pair with an appropriate HRP-conjugated secondary antibody.
  • Chromatin Immunoprecipitation (ChIP): Suitable for ChIP. Cross-link chromatin, shear, and immunoprecipitate with the antibody. Validate enrichment by qPCR at known H3K4me1 target loci.
  • Immunofluorescence (IF/ICC): Fix and permeabilize cells appropriately. Expect exclusive nuclear staining. Counterstain with DAPI or similar to confirm localization.
  • IHC-P: Antigen retrieval may be required; optimize conditions for each tissue type.
  • Dot Blot (DB) and ELISA: Ensure peptide concentration and blocking conditions are optimized to minimize non-specific binding.
