- Tissue expression: The canonical histone H3.1 (encoded by multiple genes including H3C1, H3C2, etc.) is widely expressed in somatic tissues, while the testis-specific variant H3.1t (Q16695) shows restricted expression in testicular cells.
- Lysine 27 (K27) of histone H3 is a critical residue for post-translational modifications, including citrullination PMID:16567635. - For Western blot (WB) analysis, a band at approximately 15 kDa corresponding to histone H3 is expected. Use of appropriate loading controls and histone extraction protocols may improve detection.
- In immunoprecipitation-based applications such as ChIP, ensure chromatin is sufficiently fragmented and validate antibody specificity using positive and negative genomic loci (e.g., regions with known H3K27me1 enrichment).
- Compatibility with multiple species (human, mouse, rat, and others) is predicted based on sequence homology; however, it is advisable to validate cross-reactivity in the relevant species and experimental system.
