Biological Background: MMP19 Function and Localization
- MMP19, also referred to as MMP-18 or RASI-1, is a member of the matrix metalloproteinase family that plays a critical role in extracellular matrix (ECM) remodeling.
- This endopeptidase degrades a broad range of ECM components, including aggrecan, cartilage oligomeric matrix protein (COMP), collagen type IV, laminin, nidogen, fibronectin, and type I gelatin, and is implicated in angiogenesis and neovascularization.
- The enzyme is secreted into the extracellular space and localizes to the ECM.
- MMP19 contains conserved domains typical of MMPs: a signal peptide, a propeptide with a cysteine switch motif, a catalytic domain with a zinc-binding site, and a hemopexin-like C-terminal domain; its activity is calcium- and zinc-dependent.
- Post-translational modifications include glycosylation, disulfide bond formation, and phosphorylation, and the protein exists as a zymogen requiring proteolytic activation.
- Tissue expression is widespread, with high levels in mammary gland, placenta, lung, pancreas, ovary, small intestine, spleen, thymus, prostate, testis, colon, heart, and blood vessel walls, but it is absent from brain and peripheral blood leukocytes.
