Biological Background: MICA Function and Localization

  • MICA (MHC class I polypeptide-related sequence A) is a member of the MIC family of stress-inducible proteins, also known as MIC-A or PERB11.1.
  • It acts as a ligand for the activating immunoreceptor NKG2D/KLRK1, which is expressed on virtually all human natural killer (NK) cells, γδ T cells, and CD8+ αβ T cells. Related references: PMID:11491531 PMID:11777960
  • Engagement of NKG2D by MICA stimulates NK cell-mediated cytotoxicity and cytokine production, playing a central role in innate and adaptive immune responses. Related references: PMID:10426993
  • MICA is up-regulated under conditions of cellular stress, including viral and bacterial infections, DNA damage, and heat shock, thereby serving as a danger signal for the immune system.
  • The protein is localized primarily at the cell membrane but can also be found in the cytoplasm; it is widely expressed in tissues except the central nervous system, with strong expression in gastric epithelium, keratinocytes, endothelial cells, and various epithelial tumors.
  • MICA undergoes several post-translational modifications, including N-linked glycosylation, palmitoylation, and disulfide bond formation, and contains an immunoglobulin-like domain.
  • Alternative splicing produces multiple isoforms, and the protein can be proteolytically shed from the cell surface, generating a soluble form that modulates immune responses.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein covering amino acids 24–307 (NP_000238.1), which corresponds to the extracellular domain of human MICA. This region is predicted to be exposed and may present native epitopes.
  • In Western blotting, expect a band at approximately 43 kDa under reducing conditions; however, due to glycosylation, the observed molecular weight may be slightly higher. Validation in the relevant sample system is recommended.
  • For ELISA applications, coating with the recombinant protein or using a sandwich format should be optimized; consider checkerboard titration for optimal antibody and antigen concentrations.
  • The antibody shows cross-reactivity with human, mouse, and rat, but performance in other species has not been tested and should be validated independently.
  • Standard blocking with 5% non-fat milk or BSA is commonly used for WB; if high background is observed, try adjusting the blocking agent or buffer conditions.
