Biological Background: CBX3 (HP1 gamma) Function and Localization

CBX3, also known as HP1 gamma, is a component of heterochromatin that recognizes and binds histone H3 tails methylated at Lys-9, leading to epigenetic repression. It also binds histone H1.4 methylated at Lys-26 (H1.4K26me) and is excluded from chromatin when H1.4 is simultaneously methylated and phosphorylated. Through interaction with MIS12 complex proteins, CBX3 is involved in the formation of functional kinetochores. It contributes to the conversion of local chromatin to a heterochromatin-like repressive state via H3 Lys-9 trimethylation and mediates the recruitment of methyltransferases SUV39H1/2 by the PER complex to circadian target genes such as PER2 and PER1.
