  • METAP2 encodes methionine aminopeptidase 2, also known as initiation factor 2-associated 67 kDa glycoprotein (p67, p67eIF2) and peptidase M, which plays a dual role in protein maturation and translational control.
  • It cotranslationally removes the N-terminal methionine from nascent proteins when the second residue is small and uncharged (e.g., Ala, Cys, Gly, Pro, Ser, Thr, Val). Its catalytic activity toward Met-Val peptides is over 100-fold higher than METAP1, indicating a primary responsibility for processing Met-Val and Met-Thr termini in vivo.
  • Protects eukaryotic initiation factor EIF2S1 from translation-inhibiting phosphorylation by kinases such as EIF2AK2/PKR and EIF2AK1/HCR, thereby critically regulating protein synthesis under stress conditions.

  • For Western blotting, METAP2 has a predicted molecular weight of ~53 kDa, but glycosylation or other modifications may result in slight band shifts; validate using appropriate positive controls (e.g., HeLa, HEK293, or tissue lysates) and consider using a reducing gel.
  • In ELISA applications, optimization of coating antigen concentration and antibody dilution is recommended; the antibody may capture both full-length and certain truncated/natural isoforms of METAP2.
