Biological Background: MEF2A+MEF2C Function and Localization
- Encoded by the MEF2C gene, this transcription factor belongs to the MEF2 (myocyte enhancer factor 2) family, which includes MEF2A, MEF2B, MEF2C, and MEF2D.
- Binds specifically to MEF2 elements in the regulatory regions of muscle-specific genes, activating transcription during myogenesis and cardiac morphogenesis.
- Plays a crucial role in vascular development and is involved in hippocampal-dependent learning and memory by regulating synaptic transmission.
- Essential for normal neuronal development, distribution, and electrical activity in the neocortex; may also contribute to neurogenesis and cortical architecture.
- Necessary for proper development of megakaryocytes and platelets, and for bone marrow B-lymphopoiesis; required for B-cell survival, proliferation, and IgG1 antibody responses.
- Highly expressed in brain and skeletal muscle, consistent with its roles in neurogenesis and myogenesis.
- Localizes to the nucleus, cytoplasm, and sarcoplasm; activity is modulated by alternative splicing, phosphorylation, acetylation, and ubiquitination.
- Dysregulation linked to developmental disorders and diseases including certain cardiac defects and neurological conditions (Epilepsy, Intellectual disability).
Experimental Guidance and Technical Tips
- The immunogen (aa 250-473 of MEF2C, based on Q06413) targets a C-terminal region that may detect both MEF2A and MEF2C isoforms; verify specificity in your experimental system.
- For western blotting, a band around 51 kDa is expected; consider using loading controls and account for potential isoform variants or post-translational modifications.
- Validated for immunohistochemistry (paraffin-embedded); optimize antigen retrieval methods and antibody dilution for specific tissue types.
- Suitable for ELISA applications; coat plates with recombinant antigen or peptide corresponding to the immunogen region for best results.
- Cross-reactivity with human and mouse samples is confirmed; reactivity in other species should be validated independently.
