Biological Background: MyoD Family Inhibitor (MDFI) Function and Localization
- MDFI (MyoD family inhibitor), also known as I-MF or I-mfa, is a MyoD family inhibitor that negatively regulates myogenic transcription factors by retaining them in the cytoplasm and interfering with DNA binding.
- Plays a critical role in cellular differentiation, particularly in trophoblast and chondrogenic lineages.
- Modulates Wnt signaling by binding to the axin complex and increasing free β-catenin levels, thereby affecting both canonical Wnt and JNK pathways.
- Regulates TCF7L1/TCF3 transcriptional activity by direct interaction, preventing its DNA-binding.
- Regulates the activity of mechanosensitive Piezo channels (PubMed:37590348). Related references: PMID:37590348
- Subcellular localization: predominantly cytoplasmic but can shuttle to the nucleus; presence in both nucleus and cytoplasm is noted.
- Keywords: Development protein, Differentiation, Transcription regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-160 of human MDFI, targeting the N-terminal region. Consider this when interpreting detection in isoforms or homologs.
- For Western blot, a band of approximately 25 kDa is expected. Validate using appropriate lysates from cell lines or tissues expressing MDFI (e.g., muscle, trophoblast).
- For immunofluorescence/ICC, anticipate cytoplasmic and nuclear staining patterns; include appropriate controls.
- For ELISA, the recombinant protein immunogen can serve as a standard; ensure cross-reactivity matches the species of interest.
- Cross-reactivity is reported with human, mouse, and rat; however, always verify specificity in your experimental system.
