Biological Background: MDA5 Function and Localization
- Encodes interferon-induced helicase C domain-containing protein 1 (IFIH1), also known as MDA5, Helicard, and RLR-2. It is a cytoplasmic innate immune receptor that recognizes viral dsRNA and uncapped mRNA with 2'-O-methylation deficiency.
- Upon ligand binding, MDA5 interacts with MAVS on mitochondria, triggering TBK1/IKBKE-mediated phosphorylation and activation of IRF3/IRF7, leading to type I interferon and pro-inflammatory cytokine production. Related references: PMID:28594402, PMID:32169843, PMID:33727702

- Localizes to the cytoplasm, mitochondria, and nucleus, consistent with its role in viral RNA sensing and signal transduction.

- The immunogen spans a C-terminal region (aa 900-1000) of human MDA5, which is conserved across human, mouse, and rat, supporting the reported cross-reactivity. Validate species-specific reactivity in relevant sample matrices.
- For Western blotting, the antibody may detect bands at ~117 kDa (full-length MDA5) and possibly at ~25 kDa; using a broad molecular weight range and positive controls such as interferon-treated HeLa or A549 cells is advisable.
- In ELISA, titrate the antibody against recombinant MDA5 or the immunizing peptide to determine optimal working concentrations; ensure proper blocking and washing steps.
- Since MDA5 is interferon-inducible, consider stimulating cells with IFN-alpha/beta or viral mimics (e.g., poly(I:C)) to upregulate target expression before lysis.
