Biological Background: MAVS Function and Localization
-
MAVS (Mitochondrial antiviral-signaling protein, also known as IPS-1, VISA, Cardif) is an adapter essential for innate immune defense against viruses, acting downstream of cytoplasmic RNA sensors RIG-I and MDA5 to activate NF-kappa-B and IRF3/IRF7 transcription factors, leading to induction of antiviral cytokines such as interferon-beta and RANTES. Related references: PMID:16125763 PMID:16127453 PMID:16153868
-
Following viral infection, peroxisomal MAVS initiates a rapid, interferon-independent defense, while mitochondrial MAVS triggers a delayed, interferon-dependent amplification pathway to sustain the antiviral state. Related references: PMID:20451243
-
MAVS also participates in NLRP3 inflammasome activation by mediating NLRP3 recruitment to mitochondria. Related references: PMID:23582325
-
The protein localizes to the mitochondrial outer membrane, mitochondrion, and peroxisome.
-
It is ubiquitously expressed, with highest levels in heart, skeletal muscle, liver, placenta, and peripheral blood leukocytes; at the protein level, it is detected in T-cells, monocytes, epithelial cells, and hepatocytes.
-
MAVS undergoes diverse post-translational modifications, including acetylation, phosphorylation, ubiquitination, and palmitoylation, which are critical for its signaling functions.
-
Alternative splicing generates multiple isoforms with potentially distinct regulatory roles.
Experimental Guidance and Technical Tips
- The polyclonal antibody was raised against a synthetic peptide spanning amino acids 1–100 of human MAVS, which includes the N-terminal CARD domain; verify reactivity with your target isoform if alternative splicing is a concern.
- For Western blot (WB), a band around 57 kDa is expected in human, mouse, and rat samples; consider testing in relevant virus-infected or stimulated cell lines (e.g., HEK293, HeLa, Huh7).
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), permeabilization and antigen retrieval conditions may require optimization for mitochondrial membrane proteins; including digitonin can improve mitochondrial staining.
- For ELISA applications, the antibody may be used for capture or detection, but dilution and pairing must be empirically optimized.
