Biological Background: Mast Cell Tryptase Function and Localization

  • Tryptase alpha/beta-1 (encoded by TPSAB1) is the major neutral serine protease stored in mast cell secretory granules, released upon mast cell activation and degranulation.
  • Isoform 2 (also known as tryptase beta-1) cleaves large substrates, such as fibronectin, more efficiently than isoform 1, but seems less efficient toward small substrates. Related references: PMID:18854315
  • The TPSAB1 gene produces multiple isoforms (TPS1, TPS2, TPSB1, TPSB2) through alternative splicing, with tryptase-2 being one variant.
  • Subcellular localization is predominantly secreted from mast cells into the extracellular space.
  • Tissue expression: Isoform 1 is predominant in lung, stomach, spleen, heart, and skin; isoform 2 is expressed in aorta, spleen, breast tumor, with highest levels in endothelial cells of certain blood vessels, and low levels in mast cells.

  • For IP, validate the antibody's ability to pull down the 31 kDa protein under non-denaturing conditions.
  • Because of cross-reactivity with mouse, the antibody can be used in mouse models of allergy or mast cell biology, but verify species-specific reactivity.
  • Note that tryptase may be released in soluble form; ELISA may require capture-detection optimization.
