Biological Background: MARCKSL1 Function and Localization
-
MARCKSL1 (MARCKS-like protein 1, also known as Mac-MARCKS, F52, MLP, MRP, MLP1, MACMARCKS) controls cell movement by regulating actin cytoskeleton homeostasis and the formation of filopodia and lamellipodia. Related references: PMID:22751924
-
In its unphosphorylated state, the protein promotes cell migration; upon phosphorylation by MAPK8 (JNK), it induces actin bundle stabilization, reducing actin plasticity and restricting cell movement, including neuronal migration.
-
May couple protein kinase C and calmodulin signal transduction systems.
-
Localizes to the cytoplasm, cytoskeleton, and cell membrane, consistent with its role in actin dynamics.
-
Contains actin-binding and calmodulin-binding domains, and a myristoylation site, characteristic of the MARCKS family of myristoylated alanine-rich protein kinase C substrates.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length human MARCKSL1 protein (aa 1-195), suggesting the antibody may recognize multiple epitopes; it is suitable for Western blot (WB) and ELISA applications as per supplier specifications.
- In WB, a specific band is expected at approximately 20 kDa (predicted molecular weight 19529 Da). Validate protein loading and transfer using appropriate controls.
- This antibody shows cross-reactivity with human, mouse, and rat samples; consider species-specific validation when switching model systems.
- For ELISA, the polyclonal nature may provide broad antigen capture; optimize coating and detection conditions in your assay format.
