Biological Background: MAPRE1 Function and Localization
- MAPRE1 (EB1) is a microtubule plus-end tracking protein (+TIP) that regulates microtubule dynamics by promoting cytoplasmic nucleation and elongation. Related references: PMID:12388762 PMID:16109370 PMID:19632184
- It recruits other +TIP proteins to microtubule ends via binding to a conserved Ser-X-Leu-Pro (SXLP) motif. Related references: PMID:19632184 PMID:36592928
- During mitosis, MAPRE1 positions the spindle by stabilizing astral microtubules and connecting them to the cell cortex. Related references: PMID:12388762 PMID:34608293
- It assists chromosome alignment in metaphase by recruiting the SKA complex to the spindle and stabilizing interactions with K-fibers. Related references: PMID:27225956 PMID:36592928

Experimental Guidance and Technical Tips
- For Western blot (WB), endogenous MAPRE1 migrates at ~30 kDa. Use appropriate positive controls (e.g., HeLa, HEK293). Because the immunogen spans aa 134–268, cross-reactivity with isoforms or paralogs is possible; consider validation with siRNA.
- For immunofluorescence/ICC (IF/ICC), MAPRE1 shows comet-like plus-end staining. Optimize fixation (e.g., methanol) and permeabilization to preserve microtubule structures. Co-stain with α-tubulin to confirm localization.
- This polyclonal antibody is also suitable for ELISA. Titration and assay conditions should be empirically determined for each application.
