Biological Background: MAGOH Function and Localization

  • MAGOH (Protein mago nashi homolog), also known as MAGOH1 or MAGOHA, is encoded by the MAGOH gene in humans.
  • It is required for pre-mRNA splicing as a component of the spliceosome. Related references: PMID:11991638
  • MAGOH plays a redundant role with MAGOHB as a core component of the exon junction complex (EJC) and in the nonsense-mediated decay (NMD) pathway. The EJC marks the exon-exon junction and influences downstream processes including nuclear export, subcellular localization, translation efficiency, and NMD. Related references: PMID:23917022
  • The MAGOH-RBM8A heterodimer inhibits EIF4A3 ATPase activity, stabilizing the EJC onto spliced mRNA. Interaction with PYM1 triggers EJC disassembly and enhances translation by recruiting spliced mRNAs to the ribosome.
  • MAGOH modulates splicing of BCL2L1/Bcl-X, specifically inhibiting proapoptotic isoforms such as Bcl-X(S), distinct from its canonical EJC role.
  • Subcellularly, MAGOH localizes to the nucleus (including nuclear speckles) and cytoplasm.
  • It is ubiquitously expressed and associated with mRNA processing, splicing, transport, and translation regulation; post-translational acetylation has been reported.
