Biological Background: MAFB Function and Localization
- MAFB encodes transcription factor MafB, a member of the large Maf family of basic leucine zipper (bZIP) transcription factors.
- Functions as both a transcriptional activator and repressor, depending on cellular context and target gene promoters. Related references: PMID:27181683
- Plays a pivotal role in hematopoietic lineage specification by repressing ETS1-mediated transcription of erythroid-specific genes in myeloid cells.
- Required for the differentiation of multiple cell types, including monocytes, macrophages, osteoclasts, podocytes, and pancreatic islet beta cells.
- Activates the insulin and glucagon gene promoters; together with PAX6, transactivates the glucagon gene promoter through the G1 element.
- SUMO modification controls its transcriptional activity and ability to specify macrophage fate.
- Functions either as an oncogene or a tumor suppressor, depending on the cell context.
- Localizes to the nucleus and is ubiquitously expressed across tissues.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 184–323, a central region of MAFB conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blotting, consider using whole‑cell lysates from hematopoietic cell lines (e.g., THP-1, U937) or tissues with known MAFB expression. Validate the specific band at ~36 kDa.
- For ELISA, pairing with a suitable capture/detection system may require optimization; test with recombinant MAFB protein or peptide to confirm reactivity.
- Due to MAFB’s role in differentiation, consider assessing its expression under stimulated versus unstimulated conditions to capture dynamic regulation.
