Biological Background: LAMTOR5 Function and Localization
- LAMTOR5, also known as HBXIP or XIP, is a component of the Ragulator complex, which plays a critical role in amino acid sensing and mTORC1 activation. Related references: PMID:22980980, PMID:29158492, PMID:30181260
- As part of the Ragulator, LAMTOR5 functions as a guanine nucleotide exchange factor (GEF) for the small GTPases Rag and mediates their recruitment to the lysosomal membrane. Related references: PMID:22053050, PMID:22980980, PMID:28935770
- Activated Ragulator and Rag GTPases serve as a scaffold to recruit mTORC1 to lysosomes, where mTORC1 is subsequently activated. Related references: PMID:22980980, PMID:29158492, PMID:30181260
- When complexed with BIRC5 (survivin), LAMTOR5 interferes with apoptosome assembly, preventing recruitment of pro-caspase-9 to oligomerized APAF1, thereby suppressing mitochondrial/cytochrome c-mediated apoptosis. Related references: PMID:12773388
- LAMTOR5 localizes primarily to lysosomes, but is also found in the cytoplasm and cytosol.
