Biological Background: Histone H3 Function and Localization

Histone H3 is a core component of the nucleosome, essential for DNA packaging into chromatin and for regulating DNA accessibility during transcription, repair, replication, and chromosomal stability. It forms the histone octamer together with H2A, H2B, and H4. Related references: UniProt Q16695 | UniProt P68431

Two major isoforms represented by the SWISS accessions are Histone H3.1t (Q16695), which is specifically expressed in testicular cells, and the canonical Histone H3.1 (P68431), encoded by multiple genes and widely expressed. Both share high sequence homology, including the lysine 14 residue that is the target of this antibody.

Histone H3 undergoes an extensive array of post-translational modifications (PTMs) that constitute the histone code, influencing gene expression and chromatin dynamics. These PTMs include acetylation, methylation, phosphorylation, citrullination, and lactylation. Lactylation at lysine residues has been identified at multiple positions in histone H3 (e.g., K10, K19, K24, K28, K57, K80), as reported in recent studies. Related references: PMID:31645732

The subcellular localization of Histone H3 is predominantly nuclear, where it associates with chromosomes throughout the cell cycle.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic lactylated peptide around K14 of human Histone H3. To confirm antibody specificity, consider using a non-lactylated (control) peptide or perform peptide competition assays.
  • For Western blot (WB), Histone H3 has a predicted molecular weight of approximately 15 kDa, but sample preparation and separation conditions (e.g., acid extraction or high-salt nuclear extracts) may affect detection. Validate the assay in the relevant cell or tissue system.
  • For ELISA, ensure that the lactylated antigen is properly presented. Direct or sandwich ELISA formats may be optimized using peptide or recombinant protein.
  • While the antibody is predicted to react with a wide range of species (Human, Mouse, Rat, and others) due to sequence conservation around the modification site, cross-reactivity should be experimentally confirmed for each new species or sample type.
  • Because histone lactylation is a dynamic PTM, consider including appropriate metabolic stimuli or environmental conditions to enrich for the modification prior to analysis.
