Biological Background: KRAS Function and Localization
- GTPase KRas, also known as K-Ras 2, c-K-ras, belongs to the Ras family of small GTPases.
- Functions as a signal transducer in the Ras-MAPK signaling pathway, regulating cell proliferation and survival. Related references: PMID:22711838 PMID:23698361
- Ras proteins bind GDP/GTP and possess intrinsic GTPase activity, acting as molecular switches. Related references: PMID:20949621 PMID:39809765
- Plays a role in promoting oncogenic events by inducing transcriptional silencing of tumor suppressor genes in colorectal cancer cells in a ZNF304-dependent manner. Related references: PMID:24623306
- Recognized by LZTR1, which mediates its ubiquitination by a BCR E3 ubiquitin-protein ligase complex, affecting protein stability. Related references: PMID:40934300
- Localizes to the cell membrane, endomembrane system, and cytoplasm, consistent with its role in signal transduction.
- Expressed throughout the epidermis, but weakly in the stratum corneum and dermis, suggesting a role in skin biology.
- Subject to multiple post-translational modifications including acetylation, prenylation, palmitoylation, and methylation, which regulate its membrane association and activity.
Experimental Guidance and Technical Tips
- The immunogen was designed against a region within amino acids 30-189 of human KRAS (NP_203524.1), potentially recognizing both canonical and variant isoforms. In western blotting, a band around 22 kDa is expected; consider using appropriate controls and validate in the relevant sample system.
- For immunofluorescence/immunocytochemistry, optimize fixation and permeabilization protocols; note that KRAS localizes to the cell membrane and cytoplasm, so membrane-specific staining may require careful permeabilization.
- In ELISA applications, this polyclonal antibody can be used as a capture or detection reagent; titrate to determine optimal working concentrations for your specific assay format.
- Due to cross-reactivity with mouse and rat, this antibody is suitable for studies in rodent models; however, always validate species-specific reactivity in your experimental setup.
