Biological Background: YTHDF2 Function and Localization
- YTHDF2 is an m6A reader protein that specifically binds N6-methyladenosine (m6A)-modified RNAs to regulate their stability and processing. Related references: PMID:24284625, PMID:26046440, PMID:26318451
- It promotes degradation of m6A-containing mRNAs by recruiting the CCR4-NOT deadenylase complex via direct interaction with CNOT1. Related references: PMID:27558897
- In cooperation with RIDA/HRSP12, it directs transcripts bearing a specific binding site (5'-GGUUC-3') toward endoribonucleolytic cleavage by the ribonuclease P/MRP complex. Related references: PMID:30930054

Experimental Guidance and Technical Tips
- For WB, a starting dilution of 1:1000 is often effective; however, optimization is recommended. Validate specificity using knockout cell lines or siRNA silencing.
- For IHC-P, perform heat-induced antigen retrieval and consider a dilution range of 1:50-1:200.
- For IP, typical antibody amounts of 1-2 µg per 500 µg total protein may be tested.
- For ELISA, coating with the immunogen peptide or recombinant YTHDF2 is suggested.
- Given the m6A-binding function, consider potential competition with endogenous methylated RNA in IP and other RNA-associated assays.
