XIAP is a multi-functional protein that directly inhibits apoptosis by binding and blocking the active sites of caspases-3 and -7, and by maintaining caspase-9 in an inactive monomeric state.

As an E3 ubiquitin-protein ligase, XIAP catalyzes Lys-63-linked polyubiquitination of RIPK2 downstream of NOD1/2, creating a scaffold for NF-κB and MAP kinase activation, and also ubiquitinates RIPK1, MAP3K2, and other signaling mediators to fine-tune inflammatory and survival pathways.
