Biological Background: β-Catenin Function and Localization
- β-Catenin is the key downstream effector of the canonical Wnt signaling pathway. In the absence of Wnt, it forms a destruction complex with AXIN, APC, and GSK3B, leading to its phosphorylation, ubiquitination, and proteasomal degradation. Upon Wnt activation, β-catenin accumulates and translocates to the nucleus where it acts as a coactivator for TCF/LEF transcription factors. - β-Catenin is a component of the E-cadherin:catenin adhesion complex, linking cell adhesion to the actin cytoskeleton at adherens junctions.
- It promotes epithelial to mesenchymal transition (EMT) by driving transcription of CTNNB1/TCF-target genes. Related references: PMID:29910125
- Subcellularly, β-catenin localizes to the cytoplasm, nucleus, cell membrane, adherens junctions, centrosome, spindle poles, and basal body of cilia, reflecting its diverse functions.

Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa1–100) of human β-catenin, which contains critical residues for phosphorylation-dependent degradation. Consider using the antibody to monitor total β-catenin levels versus phosphorylated forms.
- For Western blotting, the expected band is at ∼85 kDa. Validate expression in your sample system, as β-catenin is widely expressed but subject to regulated turnover.
- For immunohistochemistry (paraffin), include appropriate antigen retrieval steps and control for membrane, cytoplasmic, and nuclear localization.
- The antibody is suitable for detecting β-catenin across human, mouse, and rat. Validate cross-reactivity in your target species.
- KO validation confirms on-target recognition; however, as with any antibody, performance may vary by application and sample type—independent validation is recommended.
