Biological Background: TTLL12 Function and Localization

  • TTLL12 (Tubulin--tyrosine ligase-like protein 12), also known as dJ526I14.2, is an inactive member of the TTLL family that negatively regulates tubulin post-translational modifications, including detyrosination of the C-terminus and polyglutamylation of glutamate residues. Related references: PMID:20162578, PMID:23251473
  • It indirectly promotes histone H4 trimethylation at Lys-20 (H4K20me3), contributing to chromatin structure and epigenetic regulation. Related references: PMID:23251473
  • By controlling tubulin and histone H4 modifications, TTLL12 plays a role in mitosis and maintaining chromosome number stability. Related references: PMID:20162578, PMID:23251473
  • During RNA virus infection, TTLL12 acts as a negative regulator of the RIG-I pathway by preventing MAVS binding to TBK1 and IKBKE, thus modulating innate immune responses. Related references: PMID:28011935
  • TTLL12 localizes to the cytoplasm, midbody, centrosome, spindle, and nucleus, consistent with its roles in cell division and gene regulation.
  • It is expressed in the basal layer of prostate and endothelial cells, with increased expression observed in prostatic intraepithelial neoplasia and metastatic lesions, suggesting a potential role in cancer progression.
  • As an ATP- and nucleotide-binding protein, TTLL12 is identified through proteomics and is a reference proteome component.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (a.a. 485–644), which may confer epitope specificity; consider using full-length protein or peptide competition to confirm binding.
  • For Western blot, a band near 74 kDa is expected; validate using positive and negative controls, and consider using KO-validated lysates if available.
  • The antibody reacts with human, mouse, and rat samples, making it suitable for cross-species studies in those organisms; always verify reactivity in your model system.
  • For ELISA, titration of the antibody and optimization of coating conditions are recommended to achieve optimal signal.
  • Since the product is labeled as KO Validated, it may be useful for confirming target knockdown; however, users should perform appropriate validation experiments tailored to their specific experimental design.
