Biological Background: SMARCC1/BAF155 Function and Localization

  • SMARCC1 (also known as BAF155, SWI/SNF complex 155 kDa subunit, HYC5, Rsc8, SRG3, SWI3, CRACC1) is a core subunit of the ATP-dependent SWI/SNF chromatin remodeling complexes.
  • Involved in chromatin remodeling: it alters DNA-histone contacts within nucleosomes in an ATP-dependent manner, facilitating transcriptional activation and repression of select genes. May stimulate the ATPase activity of the catalytic subunit of the complex. Related references: PMID:10078207 | PMID:29374058
  • Component of the neural progenitor-specific npBAF complex, which is essential for self-renewal and proliferative capacity of multipotent neural stem cells. As neurons exit the cell cycle and differentiate, a switch to the neuron-specific nBAF complex occurs, involving substitution of homologous subunits (e.g., ACTL6A to ACTL6B, PHF10 to DPF1 or DPF3). The nBAF complex, along with CREST, regulates genes required for dendrite growth.
  • Localizes to both the nucleus and the cytoplasm.
  • Widely expressed across tissues, including brain, heart, muscle, placenta, lung, liver, kidney, and pancreas.
  • Post-translationally modified by acetylation, methylation, phosphorylation, and ubiquitination (isopeptide bond). These modifications may regulate its assembly into complexes or chromatin targeting.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 80–320 of human SMARCC1, a region that may be exposed and antigenic; verify specificity on endogenous protein in relevant cell or tissue lysates.
  • For WB, consider the predicted molecular weight of ~123 kDa; use reducing conditions and appropriate molecular weight markers. Validate with a loading control and, if available, KO cell lines.
  • When performing IP, optimize antibody-to-lysate ratios and include appropriate isotype controls. Crosslink the antibody to beads if heavy/light chain interference is a concern.
  • For ELISA, determine optimal coating antigen concentration and antibody dilution through checkerboard titration. Include blocking steps and appropriate blanks.
