- Protein NDRG1 (N-myc downstream-regulated gene 1), also known as DRG-1, Cap43, RTP, and Rit42, is a member of the NDRG family of stress-responsive proteins.
- NDRG1 functions as a tumor suppressor in multiple cell types and is involved in p53/TP53-mediated caspase activation and apoptosis (necessary but not sufficient).
- Plays a critical role in cell trafficking, particularly in Schwann cells, and is essential for the maintenance and development of the peripheral nerve myelin sheath.
- Required for vesicular recycling of CDH1 (E-cadherin) and transferrin (TF), potentially contributing to lipid trafficking.
- Protects cells from spindle disruption damage and functions in the p53/TP53-dependent mitotic spindle checkpoint, regulating microtubule dynamics and maintaining euploidy.
- Localizes to the cytoplasm, cytosol, centrosome, nucleus, and cell membrane, reflecting its diverse cellular roles (UniProt: Q92597).
- Ubiquitously expressed, with highest levels in placental membranes, prostate, kidney, small intestine, and ovary; also expressed in Schwann cells; reduced expression in adenocarcinomas.
