Biological Background: JAK2 Function and Localization
- Tyrosine-protein kinase JAK2 (Janus kinase 2) is a non-receptor tyrosine kinase critically involved in signaling by many cytokine receptors, including those for interleukins, interferons, and hormones.
- Upon ligand engagement, JAK2 phosphorylates specific tyrosine residues on the receptor cytoplasmic tails, creating docking sites for STAT proteins, which are then phosphorylated, dimerize, and translocate to the nucleus to activate target gene transcription.
