Biological Background: IFITM3 Function and Localization
- IFITM3 is an interferon-induced antiviral protein that restricts the entry of a broad range of enveloped viruses, including influenza A virus, SARS-CoV, SARS-CoV-2, Ebola virus, and HIV-1, by inhibiting viral fusion with cholesterol-depleted endosomes (PubMed:26354436; PubMed:33239446; PubMed:33270927). - Belongs to the dispanin family (DSPA2b) and is also known as 1-8U or IP15, with interferon-inducible expression.
- Plays a critical role in the structural stability and function of vacuolar ATPase (v-ATPase) on endosomes, facilitating proper clathrin localization and endosomal acidification required for its antiviral activity (PubMed:26354436). Related references: PMID:26354436

- The immunogen covers amino acids 1-133 of human IFITM3, which includes the N-terminal region. For Western blotting, a band around 15 kDa is expected; validate in a relevant interferon-stimulated cell line.
- For IHC-P and IF/ICC, consider optimizing antigen retrieval and permeabilization steps, as IFITM3 is an integral membrane protein requiring membrane permeabilization for detection.
- Cross-reactivity with mouse and rat IFITM3 is listed; however, sequence alignment and pilot experiments in these species are recommended to confirm antibody performance.
- This antibody is validated for ELISA; it may be suitable for sandwich ELISA development when paired with a detection antibody recognizing a non-overlapping epitope.
