- HIF1AN, also known as Factor inhibiting HIF-1 (FIH-1), is a dioxygenase that belongs to the oxidoreductase family and requires iron as a cofactor.
- Functions as an oxygen sensor: Under normoxic conditions, it hydroxylates HIF-1 alpha at asparagine 803 in the C-terminal transactivation domain, blocking interaction with transcriptional coactivators like CBP/p300.
- Involved in transcriptional repression through interactions with HIF1A, VHL, and histone deacetylases.
- Also hydroxylates specific asparagine residues within ankyrin repeat domains (ARD) of substrates such as NFKB1, NFKBIA, NOTCH1, ASB4, PPP1R12A, and others, expanding its regulatory scope.
- Additionally, it hydroxylates aspartate and histidine residues in ARDs of ANK1 and TNKS2, respectively, demonstrating versatile substrate specificity.
- Negatively regulates NOTCH1 activity, thereby accelerating myogenic differentiation.
- Positively regulates ASB4 activity, promoting vascular differentiation.
